Does Darius trigger on being the second played card and keep buffing later?

If Darius, Trifarian is my second card played in a turn, does he get +2 and ready, and if he stays in play can he gain +2 again on later turns?

Yes to both. Darius can see that he was the second card played once resolved, and while in play he continues to gain the +2 when its condition is met.
